A soprano saxophone is in B♭ and sounds a major second below what it reads — the same transposition as a trumpet or a B♭ clarinet, and one octave above where a tenor lands. Written C comes out as concert B♭4, 466.16 Hz.
The transposition is the easy part. What makes this the hardest saxophone to play in tune is physical: it is short. A tube this length gives the mouthpiece enormous leverage over the pitch and gives your embouchure more still, so the useful thing a tuner does on a soprano is not tell you where to put the cork — it is show you how much of your pitch is coming from your jaw.

Why a millimetre matters here and not on a baritone
Pitch on any wind instrument is a proportion of the air column, not an absolute distance. A millimetre off the cork of a baritone changes a very long tube by a very small fraction; the same millimetre on a soprano changes a short one by a large fraction. This is why soprano players who came from alto consistently overshoot on their first attempt — the gesture that used to be a small correction is now a large one.
The practical consequence is to move in half-millimetres and re-attack the note each time, and to accept that the final few cents are not going to come from the cork at all.
The routine
- Warm it thoroughly. Small instruments warm quickly and cool quickly, so a soprano is both faster to bring up to pitch and faster to fall off it between numbers.
- Play written C at your real dynamic, jaw relaxed. Watch for concert B♭. Take the reading from a note you have held for two full seconds, not from the attack.
- Adjust in half-millimetres. Sharp, pull off; flat, push on. Re-attack rather than sustaining through the move.
- Check both octaves of written C. If the upper is sharp against the lower, the mouthpiece is too far on regardless of what the single tuning note said.
- Play a slow scale and watch the dial without correcting. On a soprano this is the most useful minute of the whole routine: it shows you which notes your embouchure is already bending, which is information no amount of cork adjustment provides.

Everything above names the concert pitch actually in the air, which is what a tuner is for and what the rest of the ensemble hears. But it is not what is printed on your part: a B♭ soprano saxophone sounds a major second lower than it reads, so written C comes out as concert B♭.
